The Metabolic Core will conduct in vivo and ex vivo metabolic experiments on mice shipped to the Case MMPC.
This core focuses primarily on isotopic measures of intracellular chemical fluxes and energy expenditure.
|
- Energy expenditure integrated over four days by the "doubly-labeled water" method
- Rates of fatty acid, cholesterol, triglycerides, or protein synthesis measured using 2H2O
- Chronic or acute arterial and jugular vein catheterization
- Acute catheterization of portal vein, bladder, or gastric catheterization
- Insulin clamp, pancreatic clamp
- Food intake
- Body temperature
- Urine and blood chemistry analysis
- Glycosylated hemoglobin
- Liver perfusion and heart perfusion
- Tissue perfusion and fixation
Analytical and Metabolomic Core

Tissues, blood, plasma and urine from experiments done at the MMPC or at the parent lab using isotopic tracer ‘kits’ will be analyzed.
|
- 2H-and 18O-enrichment of plasma or urine for total energy expenditure
- 2H-enrichment of plasma or urine for calculating total body water
- turnover of glucose or glycerol with [6,6-2H2]glucose and [2H5]glycerol
- turnover of glucose and glycerol with [6,6-2H2]glucose and [2H5]glycerol
- profile of 120 organic acids in urine
- profile of acylcarnitines
- profile of long-chain acyl-CoAs in tissues
- profile of acetyl-CoA, propionyl-CoA, succinyl-CoA, and methylmalonyl-CoA in
- tissues
- profile of citric acid cycle and gluconeogenic intermediates in tissues
- 13C-labeling pattern of the acetyl moiety of citrate
- profile of plasma or tissue free or total fatty acids C8 -C22
- amino acid profile in plasma
- activity of acetyl-CoA carboxylase or malonyl-CoA decarboxylase in tissues
- rates of FA and cholesterol synthesis in tissues from the incorporation of 2H or 13C
- rate of protein synthesis from the incorporation of 2H-enriched water
- metabolomic profile in plasma, urine or tissue
