Leptin-mediated regulation of spontaneous physical activity in mice

The major theme of this proposal focuses on the mechanism whereby leptin increases energy expenditure in mice with a specific emphasis on two poorly-described aspects of spontaneous physical activity-associated energy expenditure; ambulatory activity and wheel running. We propose to determine whether physiological leptin replacement increases spontaneous physical activity in mouse models of hypoleptinemia and low ambulatory activity and whether these effects can be dissociated from other effects of leptin on energy balance. In addition, we will determine whether mice with differential leptin-mediated regulation of food intake and ambulatory activity can be used to identify key neuronal signals regulating spontaneous physical activity
