Description The Akt1 KO mice were made using Cre with a Nestin promoter
and enhancer, the Akt2 with Cre under the control of the
albumin promoter with an AFP enhancer, and the Foxo1 using
Cre driven by the 1.2 kb liver-specific a1-antitrypsin
promoter. These mice were then crossed with each other for
sufficient generations (>5 backcrosses) to obtain triple
knockouts.

Akt1 knockin: A targeting vector was designed to insert a
single loxP site upstream of exon 3, and a loxP-flanked
neomycin resistance (neo) cassette downstream of exon 4 of
the gene.

Akt2 knockin: An frt-flanked PGK-neomycin resistance
cassette was inserted into intron 5, and loxP sites were
inserted flanking exons 4 and 5.

Foxo1 knockin: Exon 2 was replaced with a floxed neo
cassette and a modified exon 2 with nucleotide substitutions
that result in the amino acid substitutions of arginine for
lysine at positions 219, 242, 245, 259, 262, 271, 291
(K219R, K242R, K245R, K259R, K262R, K271R, and K291R). These
mutations produce a constitutively deacetylated allele.
Cre-mediated recombination removed the neo cassette.

Nes-cre: This transgene expresses Cre recombinase with a
human growth hormone polyadenylation signal under the
control of a rat nestin (Nes) promoter and the nervous
system-specific enhancer present in the second intron. This
promoter expresses Cre recombinase in neuronal and glial
cell precursors. Cre recombinase activity is present in
nervous tissue by embryonic day 11. The transgene was
localized to chromosome 12 using FISH with a rat nestin
promoter probe (J:180046). The insertion caused a 17 bp
deletion.

Alb-cre: The transgenic construct contained an albumin
promoter and an alpha fetoprotien enhancer adjoined to
sequence encoding cre recombinase. Cre expression was driven
in the developing liver.
