Description The L1L2_Bact_P cassette was inserted at a position on
Chromosome 12 upstream of exon 7 of the gene. The cassette
is composed of an FRT site followed by lacZ sequence and a
loxP site. This first loxP site is followed by neomycin
under the control of the human beta-actin promoter, SV40
polyA, a second FRT site and a second loxP site. A third
loxP site was inserted downstream of exon 7. Flp-mediated
recombination removed the FRT-flanked neo cassette leaving
exon 7 floxed.
To generate the Adipoq-Cre BAC transgenic mice, the 245 kb
RP23-90G21 bacterial artificial chromosome (BAC) containing
at least five mouse genes was modified by replacing the
starting ATG and 222 bp of the adiponectin gene with the
starting ATG and coding sequence of a Cre recombinase gene.
This modified BAC transgene was microinjected into pronuclei
of fertilized one-cell stage FVB/NJ embryos. Transgenic mice
were identified and bred to C57BL/6J wildtype mice to
establish founder lines. A single transgenic founder line
was characterized.
