Description Liver-Rescue of Ceacam1 in Global Ceacam1 (Cc1) Knockout
(KO) Mice

Ceacam1 KO: A neomycin resistance cassette replaced the
first two exons of the gene. These exons contain the ATG and
encode the first Ig-like domain. RT-PCR confirmed absence of
transcript in mutants. Western blot analysis on colon and
liver did not detect protein. Northern blot analysis on
colon and liver did not detect transcript.

Cc1 transgene: The “minigene” construct was obtained by
subcloning 519 nucleotides of proximal APOA1 promoter 5' of
a Ceacam1 rat minigene containing intron 1, subcloned into
plasmid cytomegalovirus (pCMV)-3Tag-3A plasmid at Not1 site
to encode a FLAG-tagged rat CEACAM1 protein.

Creation Method transgenic
Phenotype Description Cecam1 KO mice represent a model for insulin resistance due
to the impairment of insulin clearance. Ceacam1 KO mice
exhibit higher body mass, higher fat mass, hyperinsulinemia,
insulin resistance, obesity and typical features of NAFLD.
Rescuing Ceacam1 exclusively in the liver prevents all these
abnormalities. Ceacam1 rescue mice are lighter than Ceacam1
KO mice and are protected against insulin resistance.
